Monograph

Sermorelin (5mg) — Research Peptide

Sermorelin (GHRH 1-29) is a synthetic 29-amino acid peptide corresponding to the biologically active N-terminal fragment of growth hormone-releasing hormone (GHRH). It retains full GHRH receptor binding activity despite being approximately two-thirds the length of native GHRH (44 amino acids), making it the most studied truncated GHRH analogue in both preclinical and clinical research.

GHRH Receptor Pharmacology

Sermorelin binds the GHRH receptor (GHRHR) on somatotroph cells of the anterior pituitary, stimulating synthesis and pulsatile release of growth hormone. Unlike exogenous GH administration, sermorelin acts upstream of GH itself, preserving the normal negative feedback loop — as GH and IGF-1 levels rise, somatostatin release increases to suppress further GHRH receptor stimulation, maintaining physiological GH pulse characteristics. This feedback-preserving mechanism distinguishes GHRH analogues from direct GH administration in research contexts.

Sermorelin has an endogenous half-life of approximately 7-11 minutes due to rapid DPP-IV cleavage and renal clearance — significantly shorter than modified analogues such as CJC-1295 DAC (~6-8 days). This short half-life makes sermorelin particularly useful in research designs requiring acute, time-limited GHRH stimulation rather than sustained receptor engagement.
