IGF-1 LR3 (Insulin-like Growth Factor-1 Long Arg3) is a synthetic analogue of native IGF-1 with a 13-amino acid N-terminal extension and arginine substitution at position 3. These modifications reduce binding affinity for IGF binding proteins (IGFBPs), extending the plasma half-life from approximately 10-15 minutes (native IGF-1) to 20-30 hours. This pharmacokinetic profile makes IGF-1 LR3 an essential research tool for studying sustained IGF-1 receptor signalling without the confounding rapid clearance of the native molecule.

IGF-1 Receptor Signalling

IGF-1 LR3 binds the IGF-1 receptor (IGF-1R) with affinity comparable to native IGF-1, activating the PI3K-Akt-mTOR pathway — the primary driver of cellular protein synthesis, survival, and proliferation. Downstream of Akt, mTOR complex 1 (mTORC1) phosphorylates S6K1 and 4E-BP1 to stimulate ribosomal biogenesis and mRNA translation. Simultaneously, Akt phosphorylates and inhibits FoxO transcription factors, suppressing the ubiquitin-proteasome muscle atrophy pathway. These combined anabolic and anti-catabolic effects make IGF-1 LR3 a key research tool in muscle biology.
